The Duquesne Family Office, the investment firm of billionaire investor Stanley Druckenmiller, took part in SonoThera’s oversubscribed $125 million Series B financing, which closed in June 2026. The round was led by Vida Ventures and drew a broad group of life-science backers, including ARK Invest, CureDuchenne Ventures, Leaps by Bayer, Otsuka Pharmaceutical, SymBiosis, UCB Ventures, Vivo Capital, ARCH Venture Partners, Johnson & Johnson Innovation, RA Capital, and Vertex Ventures HC, with Duquesne participating as an existing investor. SonoThera, based in South San Francisco, is developing a nonviral approach to genetic medicine. Rather than using viruses to carry genetic material into cells, its platform pairs standard clinical ultrasound with gas-filled microbubbles (a delivery system it calls RIPPLE) and a payload engineering technology (PORE) to move DNA and RNA into targeted tissue, with the aim of enabling the repeat dosing that conventional gene therapies cannot support. The company plans to use the capital to advance its two lead programs, one for Duchenne muscular dystrophy and one for autosomal dominant polycystic kidney disease, toward a first clinical trial expected in 2027, and to broaden its pipeline across other organ systems.

Duquesne Family Office is the private investment firm of Stanley Druckenmiller, one of the best-known figures in global macro investing. Druckenmiller founded the hedge fund Duquesne Capital Management in 1981, ran money for George Soros’s Quantum Fund between 1988 and 2000 (the pair are closely tied to the 1992 bet against the British pound), and closed Duquesne Capital in 2010 to convert it into a family office managing his own wealth. The firm is based in New York and named after a park in Druckenmiller’s native Pittsburgh. It invests across public and private markets, and its quarterly disclosures of US-listed holdings have recently run at roughly $4 billion, concentrated in a small number of high-conviction positions with a visible tilt toward healthcare, semiconductors, and AI-related names. Alongside the office, Druckenmiller is a major philanthropist through the Druckenmiller Foundation, including a $100 million gift to establish a neuroscience institute at NYU’s medical school.
